Competing antibodies bind the target where the drug normally binds (free receptor), while non-competing antibodies bind a different site (total receptor). The ratio provides the level of receptor occupancy of the drug product.

Half the sample is saturated with the drug product; the other half is not. The drug is detected via secondary antibody staining โ€” particularly useful when monoclonal antibodies to the receptor are unavailable.
